Summary; Sam and Dean are hiding out from Anna

Dean pulls the Impala up behind the building Sam is in and dials the kid's number. He sits and waits for him to come out, engine idling. He's got a couple of six packs of beer in the back of the car and half a pie that Jo brought for him. They're set to wait out termin-angel.
"Come on Sammy," he mutters and fiddles with the radio until AC/DC 'Highway to Hell' comes up.

And it's clear from the tone of his voice he does need Sam. In the same way that Sam doesn't function properly without Dean, Dean doesn't function properly without Sam. They are wholly and completely dysfunctional. The words are more there to avoid the chick flick moment than to actually cast any sort of meaning on the conversation. His tone conveys all of that.
